DEFINITION
(2S)-2-Amino-3-(1H-imidazol-4-yl)propanoic acid hydrochloride monohydrate.
Product of fermentation or of protein hydrolysis.
Content: 98.5 per cent to 101.0 per cent (dried substance).
CHARACTERS
Appearance: White or almost white, crystalline powder or colourless crystals.
Solubility: Freely soluble in water, slightly soluble in ethanol (96 per cent).

E. Dissolve 0.1 g in 7 mL of water and add 3 mL of a 200 g/L solution of sodium hydroxide. Dissolve 50 mg of sulfanilic acid in a mixture of 0.1 mL of hydrochloric acid and 10 mL of water and add 0.1 mL of sodium nitrite solution. Add the second solution to the first and mix. An orange red colour develops.
F. About 20 mg gives reaction (a) of chlorides.
Solution S: Dissolve 2.5 g in carbon dioxide-free water prepared from distilled water and dilute to 50 mL with the same solvent.
Appearance of solution: Solution S is clear and not more intensely coloured than reference solution.
pH: 3.0 to 5.0 for solution S.
Specific optical rotation: + 9.2 to + 10.6 (dried substance).
Ninhydrin-positive substances: to pass the test.
Sulfates: Maximum 300 ppm.
Ammonium: to pass the test.
Iron: Maximum 10 ppm.
Loss on drying: 7.0 per cent to 10.0 per cent, determined on 1.000 g by drying in an oven at 145-150C.
Sulfated ash: Maximum 0.1 per cent, determined on 1.0 g.

DESCRIPTION
L-Histidine Monohydrochloride occurs as white crystals or as a crystalline powder. It is soluble in water, and insoluble in alcohol and in ether. It melts with decomposition at about 250C (after drying).
Function: Nutrient.
REQUIREMENTS
Identification: The infrared absorption spectrum of the sample exhibits relative maxima at the same wavelengths as those of a typical spectrum as shown in the section on Infrared Spectra, using the same test conditions as specified therein.
Assay: Not less than 98.5% and not more than 101.5% of C6H9N3O2-HCl-H2O, calculated on the dried basis.
Lead: Not more than 5 mg/kg.
Loss on Drying: Not more than 0.3%.
Optical (Specific) Rotation [α]D20°: Between +8.5° and +10.5°, calculated on the dried basis.
Residue on Ignition: Not more than 0.1%.
